An aspartic acid residue at the active site of pepsin. The isolation and sequence of the heptapeptide.

چکیده
Pepsin reacts stoicheiometrically with the active-site-directed irreversible inhibitor N-diazoacetyl-l-phenylalanine methyl ester, with concomitant loss of all proteolytic and peptidolytic activity. The reagent esterifies a unique aspartic acid residue in pepsin, which is in the sequence:Ile-Val-Asp-Thr-Gly-Thr-Ser

Investigation of Consecutive Separating Arrangements of Bio active Compounds from Black Tea (Camellia sinensis) Residue
Every year lots of black tea (Camellia sinensis (L.) Kuntze) residue will produce in the factories. These residue are unusable whereas the bio active compounds can be extracted and used in the drag and food industries. Due to mentioned problems, this project was conducted years 2011 - 2012 with the aim to make a study on consecutive isolation of all bio active compounds from tea residu...
متن کاملSynthesis, purification, and active site mutagenesis of recombinant porcine pepsinogen.
In order to carry out studies on structure and function relationships of porcine pepsinogen using site-directed mutagenesis approaches, the cDNA of this zymogen was cloned, sequenced, expressed in Escherichia coli, and the protein refolded, and purified to homogeneity. Porcine pepsinogen cDNA, obtained from a lambda gt10 cDNA library of porcine stomach contains 1364 base pairs. It contains lead...
متن کاملCharacterization and sequencing of an active-site cysteine-containing peptide from the xylanase of a thermotolerant Streptomyces.
The kinetics of chemical modification of the xylanase from a thermotolerant Streptomyces T7 indicated the involvement of 1 mol of cysteine residue/mol of enzyme [Keskar, Srinivasan & Deshpande (1989) Biochem. J. 261, 49-55]. The chromophoric reagent N-(2,4-dinitroanilino)maleimide (DAM) reacts covalently with thiol groups of xylanase with complete inactivation. Protection against inactivation w...
متن کاملPrimary structure of porcine pepsin. III. Amino acid sequence of a cyanogen bromide fragment, CB2A, and the complete structure of porcine pepsin.
The complete amino acid sequence of porcine pepsin (EC 3.4.4.1) was constructed from the sequence of five cyanogen bromide fragments. The sequence of one of these fragments, CB2A, is reported here. The sequences of 4 other fragments are known from previous work. Porcine pepsin contains 327 residues with three structural variants. The active center aspartyl residue, which reacts with 1,2-epoxy-3...
متن کاملPhylogenetic Analysis of Hemagglutinin Gene of H9N2 Avian Influenza Viruses Isolated from Chicken in Iran in 2010-2011: Emerging of a New Subgroup
Background and Aims: Hemagglutinin (HA) protein of Avian Influenza (AI) plays an essential role in the virus pathogenicity. AI H9N2 subtype causes significant economic loss in broiler and layer in poultry farms in Iran. AI viruses have a great involvement in evolutionary changes at nucleotide and amino acid levels and vaccines could induce faster rates of such changes. Up-dated understanding of...
